Best, Audrey N. (University of Georgia, Athens), and W. J. Payne. Preliminary enzymatic events in asparagine-dependent denitrification by Pseudomonas perfectomarinus. J. Bacteriol. 89:1051-1054. 1965.-The initial events leading to denitrification by Pseudomonas perfectomarinus grown anaerobically in the presence of NO(3) (-) were investigated. Conversion of asparagine to malic acid was demonstrated. The nicotinamide adenine dinucleotide phosphate-linked malic acid dehydrogenase in this bacterium was shown to have a dependence for a divalent cation. A transhydrogenase was demonstrated. Oxaloacetic acid, a likely intermediate in conversion of malic to pyruvic acid, was rapidly decarboxylated by enzymes in cell-free extracts. We were not able to show reversible formation of malic acid with oxaloacetic acid and reduced nicotinamide adenine dinucleotide phosphate, nor of oxaloacetic acid with pyruvic acid and CO(2).

Modulation by copper of the products of nitrite respiration in Pseudomonas perfectomarinus.

A synthetic growth medium was purified with the chelator 1,5-diphenylthiocarbazone to study the effects of copper on partial reactions and product formation of nitrite respiration in Pseudomonas perfectomarinus. This organism grew anaerobically in a copper-deficient medium with nitrate or nitrite as the terminal electron acceptor. Copper-deficient cells had high activity for reduction of nitrat...

The requirement of RpoN (sigma factor sigma54) in denitrification by Pseudomonas stutzeri is indirect and restricted to the reduction of nitrite and nitric oxide.

The rpoN region of Pseudomonas stutzeri was cloned, and an rpoN null mutant was constructed. RpoN was not essential for denitrification in this bacterium but affected the expression levels and enzymatic activities of cytochrome cd1 nitrite reductase and nitric oxide reductase, whereas those of respiratory nitrate reductase and nitrous oxide reductase were comparable to wild-type levels. Since t...

Quorum sensing regulates denitrification in Pseudomonas aeruginosa PAO1.

Anaerobic growth of Pseudomonas aeruginosa PAO1 was affected by quorum sensing. Deletion of genes that produce N-acyl-l-homoserine lactone signals resulted in an increase in denitrification activity, which was repressed by exogenous signal molecules. The effect of the las quorum-sensing system was dependent on the rhl quorum-sensing system in regulating denitrification.

Cell biology and molecular basis of denitrification.

Denitrification is a distinct means of energy conservation, making use of N oxides as terminal electron acceptors for cellular bioenergetics under anaerobic, microaerophilic, and occasionally aerobic conditions. The process is an essential branch of the global N cycle, reversing dinitrogen fixation, and is associated with chemolithotrophic, phototrophic, diazotrophic, or organotrophic metabolis...

The conversion of nitrate in water to diatomic nitrogen gas by immobilized Pseudomonas stutzeri on vermiculite

Denitrification is a reduction of nitrate by heterotrophic and autotrophic bacteria that may ultimately produce molecular nitrogen (N2) through a series of intermediate nitrogen compounds.Vermiculite is a hydrous phyllosilicate mineral (Mg, Fe+2,Fe+3)3[(Al,Si)4O10](OH)2·4H2O with several layers for bacterial immobilization. The goal of this study was removal of nitrate from water with vermiculi...
